I trunked my OBI110's LinePort with a backend PBX.
I also hang the OBI's PhonePort on the PBX as an extension using SIP.

When a call comes in from PTSN,  I can pick up either from
1 - a phone on the PhonePort
2 - a SIP client connect to the PBX
3 - a phone directly connected to the PTSN line (hard line)

When there is a conversation going on via the Obi's PhonePort
I can pick up the call from a set directly connected to the PTSN line.

Is it possible to do the reverse? To interrupt and pick up a call
using the Obi/PBX when the call was initiated from the hard line?

If so, how would I go about setting it up?

The answer is maybe.

You can join the call from the OBi110 phone port.
Check EnableLINEPortBargeIn on the Phone Port configuration.
Now dial # (default to get dial tone on the line port) and you will join the call.

I assume you are sending the call from the PBX to the OBi110 via SIP and the OBi110 won't allow barge-in using this method. You have to get the call to the OBi110's phone port for this to work. I think you can do this with an extra OBi110. Connect the OBi110#2's line port to the OBi110#1's phone port with a telephone cord. Change the OBi110#2 DigitMap not to remove # when routed to the line port. Have the PBX route # to the OBi110#2.

I wanted to do this from the phone port before the barge-in option was available. Someone suggested reducing or increasing one of the voltage settings to trick the OBi110 to think the line port wasn't busy. This might work for you.
